Why the preservative stays dissolved, and why that concentration and not another

Benzyl alcohol is miscible with water only up to about 4 grams per 100 millilitres at room temperature; above that a second liquid phase separates. Preserved water uses 0.9 grams per 100 millilitres, about a quarter of saturation, so the solution is stable across the temperatures it meets and does not separate on cooling. The 0.9 figure is the concentration long shown to pass the antimicrobial effectiveness test with margin while remaining far below both the solubility limit and the level at which the compound irritates tissue.

The solubility limit

The hydroxyl group lets benzyl alcohol hydrogen-bond with water; the benzene ring resists it. The balance gives a modest solubility, around 40 grams per litre at 20 degrees, falling somewhat as temperature drops. A saturated solution turns cloudy on cooling as droplets separate. At 9 grams per litre there is no such risk within any ordinary storage range.

The figure is well established and appears in every physical property table for the compound.

Why 0.9

Preservative concentrations are chosen to pass the effectiveness test reliably with a safety margin, and then kept as low as that allows. Benzyl alcohol between about 0.9 and 2 per cent has been used in injectables for decades; 0.9 is the lower end that passes comfortably for water. Higher concentrations buy little antimicrobial benefit and more tissue irritation and assay interference.

The coincidence with the 0.9 per cent of normal saline is just that — a coincidence of two unrelated figures.

What follows

The product is a true solution with no tendency to separate, and its preservative content is fixed at manufacture and tested. This page explains the number on the label.
