An aromatic preservative absorbs where peptides are measured, and what that does to a spectrophotometer reading

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
The benzene ring of benzyl alcohol absorbs ultraviolet light, with a structured band around 250 to 260 nanometres and rising absorbance below 230. Peptides are quantified spectrophotometrically at 280 nanometres through aromatic residues or at 205 to 214 through peptide bonds, and at 0.9 per cent the preservative contributes a substantial background at the lower wavelengths. A reading on a solution made with bacteriostatic water is meaningful only against a blank of the same water.

The spectrum

Monosubstituted benzenes show a weak, fine-structured band near 255 nanometres and a much stronger band below 220. At 9 grams per litre the weak band is easily measurable and the strong band is very high. At 280 nanometres the contribution is small but not zero. The shape is the same as that of toluene and other simple aromatics.

It does not absorb in the visible, so it is colourless.

Consequences for measurement

A peptide without aromatic residues is measured near 214 nanometres, exactly where the preservative absorbs most. Without a matched blank the apparent concentration is inflated; with one, the subtraction is large and the result less precise. Chromatographic methods separate the preservative from the peptide and avoid the problem, which is one reason purity is determined by HPLC rather than by direct absorbance.

The retention time of benzyl alcohol is a known peak in any reversed-phase run on a solution made with this water.

What this does not say

Which water to use for what. It states a spectroscopic property of the preservative and its consequence for one kind of measurement.

Bacteriostatic water is supplied by REVIVE LAB UAE as a diluent for lyophilised research peptides, for laboratory research use only. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
