The claim that defined the compound, and the chemistry behind it

The early publications on BPC-157 from the Zagreb group reported that the peptide remained intact in human gastric juice for more than 24 hours, where most peptides are degraded within minutes, and this stability was part of the rationale for studying it as a gastric protective factor and for oral administration in animal models. The sequence supports the claim: pepsin prefers to cleave at aromatic and large hydrophobic residues, of which BPC-157 has almost none, and the proline triplet blocks cleavage around it. Stability in acid is a chemical property; it is not evidence of absorption or of effect.

What was measured

Incubation of the peptide in gastric juice collected from humans, with analysis by chromatography at intervals, showing the parent peak persisting. The measurement is straightforward and the finding has been repeated in the group own later work. Independent replication outside the group is limited, as for most BPC-157 findings.

The compound was described as a fragment of a larger gastric protein, which is the origin of the name body protection compound.

Why the sequence predicts it

Pepsin, the stomach protease, cuts preferentially after phenylalanine, tyrosine, tryptophan and leucine, and poorly next to proline. BPC-157 has one leucine, at position 14, and no aromatics. Its first eight residues contain four prolines. Acid alone does not hydrolyse peptide bonds at an appreciable rate at body temperature over a day. A peptide so composed would be expected to survive, and it does.

Aspartate-proline bonds are acid-labile over longer periods, and the sequence has none.

What it does not imply

That the peptide is absorbed intact, that it reaches any tissue, or that it acts after oral administration in any species beyond the rodent models in which it was tested. Stability is a necessary condition for oral activity and not a sufficient one. This page states the chemistry.
