Reversed-phase chromatography of the blend, and what purity means for each component

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
BPC-157, small and polar with four prolines and three acidic residues, has little hydrophobic surface and elutes early in a reversed-phase gradient. The thymosin fragment, longer and with more hydrophobic residues, elutes later. Between and around them lie the impurities of each — deletion sequences, deamidated forms — which are assigned to one component or the other by mass. Purity for each component is its main-peak area over the sum of its own main peak and its own impurities, so a blend certificate carries two purity figures from one run.

The separation

A standard acetonitrile gradient with trifluoroacetic acid resolves the two parents cleanly because their hydrophobicities differ substantially. Both are detected at 214 nanometres; neither has an aromatic residue that would register at 280, apart from the single phenylalanine of the full thymosin sequence if present in the fragment. The method is developed by injecting each component alone first.

Elution order is confirmed, not assumed, as the GLOW pages set out.

Assigning impurities

A deletion sequence of BPC-157 lacks one residue and elutes near its parent; a deamidated thymosin fragment elutes near its own. Mass spectrometry tells them apart. Each impurity is assigned to the component it derived from, and the two purity figures are calculated separately. The ratio of the two main peaks, corrected for their different responses at 214, also checks the 1:1 composition.

An impurity that cannot be assigned to either is reported as such.

On the certificate

Two purity percentages under one stated method, plus the composition ratio. A single purity figure for a two-component product is ambiguous and should be asked about.

The BPC-157 + TB500 blend is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for each component of the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
