The 1993 framing of BPC-157 as a gastric cytoprotective factor, and how the literature widened from there

The Zagreb group of Predrag Sikiric and colleagues reported in the early 1990s a protein isolated from human gastric juice, named body protection compound, and a synthetic 15-residue fragment of it, BPC-157, that protected the rat stomach against injury from ethanol, stress and non-steroidal drugs in the cytoprotection models of the time. The framing followed the Robert tradition of gastric cytoprotection, in which prostaglandins and other agents prevented mucosal damage without reducing acid. From that base the same group tested the peptide in models of the gut, then of tendon, ligament, muscle, nerve, vessel and brain injury, producing the broad single-source literature that exists today.

The cytoprotection tradition

André Robert showed in 1979 that prostaglandins protected the gastric mucosa against necrotising agents in rats, and the term cytoprotection entered gastric physiology. Through the 1980s many agents were tested in the same models — ethanol, aspirin, restraint stress — and BPC-157 was introduced into that framework in the 1990s as a native gastric peptide with the same property.

The models are standardised and reproducible in rats, which is partly why the literature grew so fast.

The widening

From stomach to the whole gut — colitis, fistulas, anastomoses — and then outward. The unifying hypothesis the group proposed was an effect on blood vessels and on the nitric oxide system that would explain protection in many tissues. Each new model produced a positive report. By the 2010s the published count ran to hundreds of papers, nearly all from Zagreb.

The page assessing the literature sets out what that concentration means for a reader.

Why it is here

Because the name and the origin frame the compound, and a reader should know where the claims began. It describes history and no use of the pack.
