Measuring the peptide directly rather than by difference

Amino acid analysis hydrolyses the peptide to its constituent amino acids with strong acid and heat, then separates and quantifies them against standards. For GHK the three amino acids glycine, histidine and lysine appear in a 1:1:1 ratio, and their total gives the amount of peptide in the sample directly, independent of water and counterion. It is the reference method for peptide content.

The procedure

A weighed sample is hydrolysed, typically in hydrochloric acid at elevated temperature, breaking every peptide bond. The free amino acids are derivatised or detected directly after chromatographic separation, and each is quantified against a calibration standard. The sum, corrected for any residues destroyed by hydrolysis, is the peptide content.

Tryptophan is destroyed by acid hydrolysis and needs separate handling; GHK has none, which simplifies the analysis.

Why it is direct

Calculating content by difference assumes everything that is not water or counterion is peptide. Amino acid analysis measures the peptide itself. The two should agree; where they do not, something unaccounted for is present. It also confirms composition — a 1:1:1 ratio of the three residues is a second identity check.

It is more labour than the difference method, which is why certificates often report the latter.

The copper

Hydrolysis releases the copper as free ions, which do not interfere with amino acid quantification but are not measured by it. Copper content is a separate elemental determination.

GHK-Cu is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
