An impurity that has the right mass and the wrong shape

Natural peptides are built from L-amino acids. During synthesis a residue can racemise to its D-form, giving a diastereomer of the target with exactly the same mass and nearly the same chromatographic behaviour. Mass spectrometry cannot see it. Chiral purity is checked by hydrolysing the peptide and analysing the amino acids on a chiral column, or by methods that resolve the diastereomers intact.

Why mass spectrometry misses it

A D-histidine has the same atoms and the same mass as L-histidine. A GHK with one D-residue is therefore indistinguishable from the correct peptide by mass alone, and on a standard reversed-phase column the two often co-elute or nearly so. A certificate reporting identity by mass and purity by HPLC has not addressed it.

Histidine is among the residues most prone to racemisation during coupling.

How it is measured

After hydrolysis, the amino acids are separated on a chiral stationary phase that resolves L from D, and the D-fraction of each is quantified. Alternatively, methods using chiral derivatising agents or specialised columns resolve the intact diastereomers. Both report a percentage of the wrong-handed form.

It is a specialised assay and not always on a routine certificate.

Why it matters for a metal complex

Coordination geometry depends on the three-dimensional arrangement of the donor atoms. A D-histidine presents its imidazole differently, so a diastereomer may bind copper differently or more weakly. For GHK-Cu, chiral purity bears on the chemistry, not only on nominal identity.

GHK-Cu is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
