How a certificate shows the complex is fully loaded

Copper content is measured by elemental methods — inductively coupled plasma optical emission or mass spectrometry, or atomic absorption — on a digested or dissolved sample. The result, as a percentage by mass, is compared with the theoretical value for the 1:1 complex. A match indicates full loading; a low value indicates copper-free peptide; a high value indicates excess copper.

The methods

Inductively coupled plasma techniques atomise the sample in a plasma and measure copper by its emission lines or its mass. Atomic absorption measures light absorbed by copper atoms in a flame or furnace. All are standard, sensitive and specific to the element, and none cares whether the copper was bound.

That last point is what makes them complementary to mass spectrometry of the intact complex.

The comparison

For a complex of about 400 daltons containing one copper of 63.5, the theoretical copper content is around 16 per cent by mass of the anhydrous complex. The measured value is compared against that, corrected for water and counterion content from the same certificate.

The arithmetic is simple; the correction for what else is in the solid is where care is needed.

Why it belongs on a certificate

Because identity by mass shows the complex is present and purity by HPLC shows the peptide is clean, but neither shows that all the peptide carries copper. Copper content closes that gap. It is part of lot documentation for GHK-Cu, available on request.

GHK-Cu is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
