The histidine that holds the copper also sets the pH behaviour

Free histidine imidazole has a pKa near 6, so it buffers around mildly acidic pH. In GHK-Cu the imidazole nitrogen is coordinated to copper and no longer behaves as a simple buffer, while the backbone amide that also coordinates has its pKa lowered by many units. Below roughly pH 5 the imidazole protonates and copper is released; above neutral the complex is stable. The pH behaviour of a GHK-Cu solution follows from this.

The free imidazole

Histidine is the only common amino acid with a side chain pKa near physiological pH, which is why it buffers biologically. In free GHK the imidazole would accept a proton at mildly acidic pH and release it as pH rises.

This is textbook amino acid chemistry.

What coordination does

Binding copper occupies the imidazole nitrogen and stabilises the deprotonated form, so the effective pKa shifts down. Copper also promotes deprotonation of the adjacent amide nitrogen, normally impossible near neutral pH, which is how the three-nitrogen chelate forms. Lowering pH reverses both: protons compete with copper and win below a threshold.

That threshold is the acid limit of the complex.

The solution

A GHK-Cu solution without added buffer settles at a pH set by the complex, its counterion and the water, and holds it weakly. Where the pH ends up determines whether the copper stays bound. This describes the chemistry; it specifies nothing about preparation.

GHK-Cu is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
