Three sets of deletion sequences in one vial

Solid-phase synthesis leaves deletion sequences, truncations and incompletely deprotected residues close in structure to the target. GHK-Cu, TB-500 and BPC-157 were each synthesised separately and each carries its own set. A blend therefore holds three impurity profiles at once, and a stated purity of, say, 98 per cent says nothing about which compound the remaining 2 per cent belongs to.

A tripeptide synthesis has few steps and few deletion products; a 17-residue and a 15-residue synthesis have more. The copper complex adds the possibility of copper-free GHK as a related species. Each profile is characteristic of its sequence and its process.

Combining them does not average them; it sums them.

What a single purity figure hides

Purity by HPLC is the main peak share of total area — but a blend has three main peaks. A certificate reporting one percentage has either summed three compounds against their impurities, which conflates them, or reported one and ignored two. Only a per-component statement describes the material.

Residual trifluoroacetate from three cleavages is also present unless exchanged, and affects how much active peptide a stated mass represents.

Why it matters in a blend especially

Deletion sequences of a receptor-active peptide may themselves be active or antagonistic. In a single vial that is one unknown; in a blend it is three, present in every experiment. Lot documentation for GLOW states per-component results for this reason, and is available on request.
