A sequence that barely changes between mammals, and what that means for the literature

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
IGF-1 is among the most conserved peptide hormones in mammals: human and rodent sequences differ at a handful of positions out of 70, and the receptor-binding surface is essentially identical. Human IGF-1, and the LR3 analogue built on it, therefore act fully on rodent, bovine and other mammalian receptors. Growth hormone, by contrast, is species-restricted in one direction because of a single salt bridge at the receptor interface. The conservation of IGF-1 is why rodent studies of the human analogue are pharmacologically meaningful, and why the Adelaide rat work used the human-sequence molecule.

The conservation

The insulin family is ancient and its receptor-binding chemistry tightly constrained; mutations that alter binding are selected against. Across mammals the IGF-1 sequence varies mainly at positions away from the receptor and binding-protein surfaces. Even between humans and chickens the identity is high. Insulin itself is similarly conserved, which is why animal insulin treated human diabetes for sixty years.

IGF-2 is comparably conserved.

The contrast with growth hormone

Growth hormone evolved rapidly in the primate lineage, and the primate receptor acquired a residue that excludes non-primate hormone. The hormone and receptor co-evolved into a species-specific pair. IGF-1 and its receptor did not, so the same ligand serves across the class. The two hormones, one step apart in the axis, have opposite evolutionary histories.

This is why the pituitary-extraction era was necessary for growth hormone and never arose for IGF-1.

For reading the literature

Rodent results with human IGF-1 or its analogues reflect the human receptor pharmacology faithfully in a way rodent results with human growth hormone also do, but for a different reason — human growth hormone happens to act on rodent receptors while rodent growth hormone does not act on human ones. Species of ligand matters less for IGF-1 than for almost any other hormone.
