The transcription factors that IGF-1 signalling switches off, and why they tie growth to ageing

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Activation of the IGF-1 receptor recruits insulin receptor substrates, activates phosphoinositide 3-kinase and then Akt, and Akt phosphorylates the FOXO family of transcription factors, causing them to leave the nucleus. In the nucleus FOXO proteins drive genes for stress resistance, DNA repair, autophagy and metabolic conservation; excluded, those programmes are turned down and growth programmes run. The worm FOXO, daf-16, is required for the longevity of daf-2 mutants, and variants in human FOXO3 are among the most replicated genetic associations with longevity. The pathway is the mechanistic link between IGF-1 signalling and ageing.

The switch

FOXO1, FOXO3, FOXO4 and FOXO6 in mammals; one, daf-16, in worms. Akt phosphorylation creates binding sites for 14-3-3 proteins that hold FOXO in the cytoplasm. When IGF-1 or insulin signalling falls, FOXO is dephosphorylated, returns to the nucleus and activates its targets. It is a binary-looking switch with graded reality: partial signalling gives partial exclusion.

Other inputs — AMPK, oxidative stress — also regulate FOXO, so it integrates several signals.

The targets

Antioxidant enzymes such as manganese superoxide dismutase and catalase; DNA repair genes; autophagy genes; the muscle atrophy genes atrogin-1 and MuRF1; gluconeogenic and lipid genes in liver; IGFBP-1. The last is why insulin suppresses IGFBP-1 and the atrophy genes are why IGF-1 preserves muscle: both are FOXO targets turned off by the same pathway.

The muscle maintenance and the longevity effects run through the same factor in opposite directions.

Why it is here

Because it is the molecular explanation for the trade-off the longevity page describes and for the muscle effects the Musarò page describes. It is pathway biology, stated as such.
