The amino-terminal tail of Long R3 IGF-1 is a legacy of how it was first made

Long R3 IGF-1 was described by Francis and colleagues in the Journal of Molecular Endocrinology in 1992, from the Adelaide group that became GroPep. IGF-1 expressed in E. coli was made as a fusion with a short leader to improve yield and recovery; the 13-residue extension is that leader, left in place because it did not impair activity and reduced binding to the IGF-binding proteins. The arginine for glutamate substitution at position 3 was added for the same reason. The extension is therefore an engineering artefact that was found to be useful, not a designed domain.

The expression problem

Small cysteine-rich proteins express poorly in bacteria on their own and are often degraded. Fusing them to a short peptide or a larger partner stabilises them and increases yield. The Adelaide work used a short leader and found that the fusion product, after refolding, was itself active.

Removing the leader would have added a cleavage step and cost yield; keeping it gave a product with a property of its own.

Why it was kept

The binding proteins contact IGF-1 near its amino terminus. An extension there, together with the arginine at position 3, lowers binding-protein affinity substantially while leaving receptor binding largely intact. In cell culture, where binding proteins secreted by the cells would otherwise sequester added IGF-1, that made the analogue more potent per unit added than the native protein.

This is the whole rationale for the molecule and the reason it became a standard media supplement.

What this means for identity

The product is defined by 83 residues, with the extension sequence and the arginine both part of the expected mass. A certificate confirming intact mass confirms both. Native IGF-1, at 70 residues, is a different molecule with a different mass and is not what the pack contains.
