The transgenic and gene-transfer mice that made local IGF-1 the model for muscle maintenance

Barton-Davis, Sweeney and colleagues reported in the Proceedings of the National Academy of Sciences in 1998 that a viral vector delivering IGF-1 into the muscles of mice increased muscle mass and strength and prevented the loss of both with age. Musarò, Rosenthal and colleagues reported in Nature Genetics in 2001 transgenic mice expressing a locally acting IGF-1 isoform under a muscle-specific promoter: muscle hypertrophy, preserved regenerative capacity in old animals, and no rise in circulating IGF-1. Both studies showed that IGF-1 acting within muscle, not from the blood, drives muscle growth and maintenance, and both are foundational to the muscle IGF-1 literature.

The gene-transfer study

An adeno-associated virus carrying the IGF-1 gene was injected into the muscles of young and old mice. Injected muscles were about 15 per cent larger and stronger in young animals, and in old animals the usual decline in mass and strength was prevented, with the treated muscles matching those of young mice. Circulating IGF-1 was unchanged, so the effect was local.

The authors noted the implications for muscle disease and for ageing, and the study was widely cited in both contexts.

The transgenic study

The mIGF-1 construct used a muscle-specific promoter and an isoform that stays in the tissue. Mice showed hypertrophy from youth, maintained it into old age, and regenerated muscle after injury better than controls. Serum IGF-1 was normal, and the mice did not show the systemic effects of circulating excess. The same construct later protected muscle in a mouse model of muscular dystrophy.

It is the clearest demonstration that where IGF-1 acts determines what it does.

What follows and what does not

That IGF-1 in muscle is anabolic and protective in mice when expressed there continuously by the muscle itself. That is not evidence about an analogue delivered from outside, which reaches every tissue and does not stay local. The pack is a research material.
