MOTS-c was the second such peptide found; the first came in 2001 and six more in 2016

Humanin, a 24-residue peptide encoded in the mitochondrial 16S ribosomal RNA gene, was reported by Hashimoto and colleagues in 2001 in the Proceedings of the National Academy of Sciences, found in a screen for factors protecting neurons. MOTS-c, encoded in the 12S gene, followed in 2015. Cobb and colleagues reported six small humanin-like peptides, SHLP1 to SHLP6, also from the 16S gene, in 2016. The class is defined by origin: short peptides encoded by open reading frames within mitochondrial DNA, with metabolic and cytoprotective activities reported mainly in cells and mice.

Humanin

Found by screening a complementary DNA library from surviving tissue in a brain affected by Alzheimer disease. Its frame lies in the 16S ribosomal RNA gene, and like MOTS-c it is read differently by the two codes — 24 residues cytoplasmic, 21 mitochondrial. It binds several receptors and has been studied for cytoprotection and metabolic effects for two decades.

It is the founding member and the most extensively studied.

The SHLPs

Six peptides of 20 to 38 residues from the 16S gene, reported in Aging in 2016, with differing and in some cases opposing effects on cell survival and metabolism in culture. They broadened the class and suggested mitochondrial DNA carries more coding capacity than its thirteen canonical proteins.

They are less studied than humanin or MOTS-c.

What the class shares

Mitochondrial genomic origin, small size, detection in circulation, decline with age in the reports that have measured it, and a literature dominated by preclinical work. MOTS-c is the member with the clearest metabolic mechanism proposed and the only one with human exercise data.

MOTS-c is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
