Six of sixteen residues are hydrophobic, and that shapes how the peptide behaves in water and on a column

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
MOTS-c carries six strongly hydrophobic side chains among its sixteen residues, clustered in the middle of the sequence, with its charged residues at the two ends. This gives it an amphipathic character: it dissolves in water by virtue of its four positive charges, but less readily than a polar peptide such as GHK-Cu, and it has a tendency to associate with itself and with surfaces. On reversed-phase columns it elutes late, among the more hydrophobic peptides in the catalogue.

Solubility

The charged termini pull the peptide into water; the hydrophobic core resists. The net result is a peptide that dissolves but may do so slowly and may show faint opalescence at high concentration, and whose solubility depends on pH and ionic strength more than a fully polar peptide would. Acidic conditions, which protonate all basic groups, favour dissolution.

The salt form matters: trifluoroacetate salts of hydrophobic peptides are often more soluble than acetate salts.

Aggregation and adsorption

Amphipathic peptides can align their hydrophobic faces and associate, and they adsorb to plastic and glass surfaces, which at low concentration can remove a meaningful fraction from solution. Both effects are recognised in peptide handling literature and are properties of the molecule rather than faults of a lot.

The 2015 paper noted membrane association as a possible feature of the peptide.

Chromatography

Late elution on reversed phase, well resolved from polar impurities and from the earlier-eluting oxidised forms, which are more polar than the parent. The hydrophobicity that complicates handling simplifies analysis.
