A mitochondrial DNA variant that changes one MOTS-c residue, and what population studies found

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
The mitochondrial DNA variant m.1382A>C, within the MOTS-c reading frame of the 12S ribosomal RNA gene, changes codon 14 from lysine to glutamine. It occurs in a substantial minority of people of East Asian ancestry and is rare in other populations. Fuku and colleagues reported in Aging Cell in 2015 that it was enriched in Japanese centenarians and proposed MOTS-c as a factor in exceptional longevity. Zempo and colleagues reported in Aging in 2021 that male carriers had higher rates of type 2 diabetes, with the association modified by physical activity. These are observational associations.

The variant

A single nucleotide change that is silent for the ribosomal RNA function of the gene but alters the peptide encoded in the overlapping frame. Lysine 14 is one of the four basic residues of MOTS-c; glutamine is neutral, so the variant peptide carries one less positive charge. In vitro work in the 2021 study suggested the variant peptide was less active in some assays.

It is one of the clearest cases of a mitochondrial DNA variant affecting a mitochondrial-derived peptide rather than a canonical protein.

The two population findings

The 2015 study compared centenarians with controls in Japan and found the variant more common in the long-lived group, within a haplogroup already associated with longevity. The 2021 study, in Japanese men, found more diabetes among carriers, less so among those who were physically active. The two findings are not contradictory — a variant could associate with both — but neither establishes mechanism.

Both studies were in Japanese populations, where the variant is common enough to study.

What follows

That natural variation in the MOTS-c sequence exists, is population-specific, and shows associations with metabolic and ageing phenotypes in observational data. The product in this catalogue is the reference sequence with lysine at 14.

MOTS-c is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
