Glucose restriction, oxidative stress and exercise each raise MOTS-c, and the pattern suggests what it is for

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
In cultured cells, MOTS-c increases under glucose restriction, serum deprivation and oxidative stress, and the 2018 work showed that these conditions also drive it from mitochondria to the nucleus in an AMPK-dependent manner. In human muscle it rises during exercise, itself a metabolic stress. The pattern is that of a signal produced when energy is short or the cell is under oxidative load, consistent with the proposal that mitochondrial-derived peptides inform the nucleus of mitochondrial state. How the mitochondrion increases production — transcription, export or translation of the frame — is not resolved.

The observations

Western blots and immunofluorescence in cell lines show more MOTS-c after hours of low glucose or after an oxidant challenge, with nuclear accumulation. Muscle biopsies in the 2021 human study show more during exercise. Plasma levels have been reported to change in metabolic disease states in both directions in different cohorts. The cell data are the most consistent.

All depend on antibody detection, with the caveats on the measurement page.

What it implies

A peptide that rises with energy stress and activates AMPK is behaving like part of the cellular energy-sensing machinery, and one that then enters the nucleus and alters stress-response genes is behaving like a retrograde signal. Both roles are plausible for a mitochondrially encoded molecule and both are proposed in the literature. The regulation is evidence for function in a way that sequence conservation alone is not.

The age-related decline would then represent a weakening of this signal.

Why it is here

Because regulation tells a reader what a molecule is for in a way that effects of administration do not. It describes cell and muscle observations and no use of the pack.
