The reading frame is conserved but not identical, and the studies used the human peptide in mice

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
The 12S ribosomal RNA gene is conserved across mammals, and the MOTS-c reading frame is present in the mouse genome, but the encoded peptide differs from the human sequence at several of its sixteen positions. The 2015 and 2021 mouse studies administered the human peptide, as most such studies do, so the results describe the human peptide acting in a mouse rather than the mouse peptide in its own species. Endogenous mouse MOTS-c, measured in those studies by antibodies raised against the human peptide, is detected to the extent the antibodies cross-react. Both facts are standard caveats in peptide research and are stated here as such.

The conservation

Ribosomal RNA genes change slowly because the RNA must fold correctly. A reading frame embedded in one is constrained by the RNA structure rather than by the peptide, so conservation of the frame does not by itself indicate selection for the peptide. The Lee laboratory and others have argued from conservation of the frame across species that the peptide is functional; the argument is suggestive rather than conclusive.

Similar reasoning applies to humanin.

Why the human peptide was used

Because it is the molecule of interest, because it is what a company would develop, and because it is what the antibodies and assays were built for. Human IGF-1 acts in mice because the receptor is conserved; whether human MOTS-c acts in mice as mouse MOTS-c would depends on a mechanism that has no identified receptor, so the question cannot yet be answered.

The results stand as results about the human peptide in mice.

Why it is here

Because it is a caveat a careful reader of the mouse literature should hold, and because it is the kind of fact this site states. It describes no use of the pack.
