MOTS-c was described by Changhan Lee, Pinchas Cohen and colleagues in Cell Metabolism in 2015. The paper identified a peptide encoded by a short open reading frame in the mitochondrial 12S rRNA region and reported effects on the folate-methionine cycle and AMPK activation in metabolic models.

The finding, in order

The work identified the reading frame, established that a corresponding peptide could be detected, and then examined what it did in cell and animal metabolic models. Those are three separate claims, and the strength of the evidence is not uniform across them.

The metabolic strand centred on the folate-methionine cycle — a one-carbon metabolism pathway — and on downstream AMPK activation. That ordering matters: the proposal is that the peptide acts on a metabolic pathway and AMPK responds, rather than the peptide acting on AMPK directly.

Why it drew attention beyond metabolism

A peptide encoded in mitochondrial DNA and acting elsewhere in the cell is a candidate messenger from mitochondrion to nucleus. That is a structural claim about how cells are organised, and it is of interest independently of anything metabolic.

Humanin, described in 2001 from the 16S region, had already established that the mitochondrial genome encoded more than the respiratory-chain components attributed to it. MOTS-c extended that to a different region of the genome.

What has not followed

There is no body of controlled human trial evidence. The literature remains preclinical and cell-based, and the open questions — how widely these peptides act, how they are regulated, how much of normal physiology they account for — are still open.
