What Does the Copper Do in GHK-Cu?
Short answer: The tripeptide binds a single copper(II) ion tightly, using the nitrogen of its amino terminus, the deprotonated nitrogen of the glycine-histidine peptide bond and the imidazole ring of histidine. The complex is the blue substance in the vial; GHK without copper is white. In the literature the copper is treated as part of the active molecule — the peptide is described as a copper carrier — rather than as an additive.
GHK has an affinity for copper comparable to that of albumin, which is why the complex forms readily in plasma and why the two are usually studied together.
Copper itself is a cofactor for enzymes involved in collagen cross-linking, which is one of the mechanisms proposed for the effects reported in culture; the chemistry is better established than the biology.
