Participants were assigned to placebo or to one of several dose groups, with the higher groups reaching their target through staged escalation.
The primary endpoint was percentage change from baseline body weight at 24 weeks. Change at 48 weeks, and the proportions reaching reductions of 5, 10 and 15 per cent, were secondary endpoints.
Weight reduction was dose-dependent. At 48 weeks the highest dose group recorded a mean reduction of 24.2 per cent of body weight against 2.1 per cent for placebo, and reductions in that group had not plateaued by the end of the trial period. Lower dose groups fell between those figures in order.
The paper also reported changes in waist circumference, blood pressure, glycated haemoglobin, fasting glucose and lipids as secondary measures. Gastrointestinal adverse events were the most common and were dose-related, mostly mild to moderate, and most frequent during escalation.
It was a phase 2 trial of 338 people over 48 weeks. It was not powered for cardiovascular outcomes, it did not study maintenance after stopping, and its authors describe it as supporting progression to phase 3 rather than as establishing efficacy or safety for use.
