The two chemical weak points of the GHRH sequence, known from decades of work on the native hormone

Stability studies on growth hormone-releasing hormone and its 1-29 fragment through the 1980s and 1990s identified two principal degradation routes. Asparagine 8, followed by serine, deamidates through a succinimide intermediate to aspartate and isoaspartate, the latter in the majority; the isoaspartyl form binds the receptor poorly. Methionine 27 oxidises to the sulfoxide in the presence of oxygen or light, and the oxidised peptide has reduced activity. Tesamorelin retains both residues and both routes. Deamidation needs water and proceeds fastest near neutral pH; both reactions are slow in the dry solid, which is why the product is lyophilised and stored cold and dark.

Deamidation

The asparagine side chain amide attacks the backbone nitrogen of the following residue, forming a five-membered succinimide ring that hydrolyses to a mixture of aspartate and isoaspartate, the latter with the backbone rerouted through the side chain. For GHRH the Asn-Ser sequence at positions 8 and 9 is the vulnerable site. The reaction adds one dalton and a negative charge, and the isoaspartyl product separates on reversed-phase and ion-exchange chromatography.

Deamidated GHRH has been reported to retain a fraction of the activity of the intact peptide.

Oxidation

Methionine 27 lies in the carboxyl-terminal region, outside the minimal 1-29 active core but near its end. Oxidation to the sulfoxide adds sixteen daltons and is driven by dissolved oxygen, peroxide traces and light. The sulfoxide elutes earlier on reversed phase. The hexenoyl group of tesamorelin is a six-carbon alkene and is itself a potential oxidation site, though it is not reported as a major degradant.

Protect-from-light storage addresses the photochemical route.

On the certificate

Both degradants appear within related substances under the stated chromatographic method. A fresh lot shows little of either; a solution left in air and light shows more. The batch certificate for the pack reports what was found.

Tesamorelin is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
