Mice without the releasing hormone, and what their phenotype confirmed about the axis

Alba and Salvatori reported in 2004 a mouse in which the GHRH gene had been disrupted. The animals were born normal in size and became proportionate dwarfs from about two weeks of age, with a hypoplastic anterior pituitary containing few somatotrophs, very low growth hormone and IGF-1, and normal fertility; they grew when given growth hormone. The phenotype matched the little mouse, which lacks the receptor, and confirmed that the releasing hormone is required both for somatotroph expansion during development and for growth hormone secretion after it. Both models define the one situation in which a releasing-hormone analogue cannot act in the receptor case, and the one in which it restores the missing signal in the ligand case.

The phenotype

Somatotrophs are present at birth but fail to proliferate, so the pituitary stays small; growth hormone is nearly undetectable; IGF-1 is low; growth falls behind after weaning and adult size is about half of normal. Unlike mice lacking the pituitary transcription factors, other pituitary cell types are intact. The mice are long-lived relative to wild type, as other growth-hormone-deficient mice are.

Giving growth hormone restores growth, placing the defect upstream of the pituitary response.

What it confirms

That GHRH is necessary and not redundant — ghrelin and other secretagogues do not compensate for its absence; that the developmental expansion of somatotrophs depends on it; and that the receptor and ligand knockouts match, so the receptor has no other important ligand. For a releasing-hormone analogue the models say what it needs: an intact receptor and somatotrophs, and in that setting it supplies the signal the ligand knockout lacks.

Human GHRH deficiency without receptor mutation is rare and poorly characterised, so the mouse is the main model.

Why it is here

Because the knockout is the genetic proof of the pharmacology on which tesamorelin rests. It describes mouse genetics and no use of the pack.

Tesamorelin is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
