How tesamorelin is made, where the hexenoyl group goes on, and why a 44-mer is harder than a 15-mer

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
The peptide is assembled on a resin that releases a carboxyl-terminal amide, matching the amidated leucine 44 of the natural hormone. Each cycle removes the amino-protecting group from the growing chain and couples the next protected amino acid; forty-three couplings build the chain from leucine 44 back to tyrosine 1. The trans-3-hexenoic acid is then coupled to the free amino terminus of tyrosine 1 in the same way as an amino acid. Acid cleavage releases the peptide and removes the side-chain protecting groups; preparative chromatography separates it from deletion sequences and other by-products; ion exchange converts it to the acetate; lyophilisation gives the solid. Overall yield of a 44-mer is a small fraction of theoretical, which is reflected in its price relative to shorter peptides.

Chain assembly

Each coupling is slightly incomplete, and the fraction of chains missing a residue compounds over forty-three cycles; a 99 per cent coupling efficiency gives about 65 per cent full-length chains, a 98 per cent efficiency about 42 per cent. Long hydrophobic stretches can aggregate on the resin and slow coupling further, so difficult sequences get double couplings and specialised solvents. The arginines, with bulky protecting groups, and the methionine, prone to oxidation, each add handling constraints.

This is why a 44-mer costs several times a 15-mer per milligram.

The acyl group and the amide

Coupling a fatty acid to the amino terminus uses the same chemistry as an amino acid coupling and goes to completion easily; the position is unambiguous because only one free amine remains. The carboxyl-terminal amide comes from the resin linker and is confirmed by the mass, which is one dalton lower than the free acid would be.

Both features are checked on the certificate by mass.

Why it is here

Because how the molecule is made determines what its impurities are and what its price reflects. It describes manufacturing and no use of the pack.

Tesamorelin is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
