Fifteen years from a thymus extract to a 43-residue peptide, and a surprise about where it came from

Goldstein, White and colleagues reported in 1966 a partially purified extract of calf thymus, thymosin fraction 5, that restored immune function in thymectomised animals, and set out to isolate the active components. Thymosin beta-4 was purified from the fraction and sequenced by Low, Hu and Goldstein in 1981, a 43-residue acetylated peptide. Within a few years it was found in high concentration in platelets, macrophages and most other cells and shown to bind actin, which reclassified it from a thymic hormone to a ubiquitous intracellular actin-sequestering protein that also appears outside cells.

The thymic era

The 1960s established the thymus as the organ that educates T cells, and the search for a thymic hormone followed. Fraction 5 was a mixture of dozens of peptides with immune activity in animal assays, and the alpha and beta thymosins were named for their position on isoelectric focusing of it. Thymosin alpha-1, also from the fraction, did become an immunomodulatory medicine in some countries.

Beta-4 was the most abundant peptide in the fraction.

The reclassification

Through the 1980s thymosin beta-4 was found at micromolar concentrations in cells with no connection to the thymus and identified as the main sequestering protein for monomeric actin — Safer, Nachmias and colleagues established the actin binding around 1990. Its presence in thymus extract was because it is in every tissue. The name stayed; the hormone concept did not.

This history is why the compound is sometimes misdescribed as an immune peptide.

What came after

The extracellular activities — wound healing in animals from 1999, cardiac repair in mice from 2004, eye trials in humans from the 2010s — are a second literature built on the full protein. The fragment in this pack is a third thing again, described on its own pages.
