IGF-1 LR3: the two changes in the name

Published 24 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
LR3 describes an 83-amino-acid analogue of IGF-1 carrying a 13-residue N-terminal extension (the "Long") and an arginine substituted at position 3 (the "R3"). Both changes reduce binding to IGF-binding proteins.

The problem the analogue was built for

Native IGF-1 in circulation is largely bound to IGF-binding proteins, which control how much is free to reach receptors. In culture, binding proteins secreted by the cells themselves do the same thing — they absorb added IGF-1 and make the delivered amount hard to know.

That is a measurement problem: the concentration added and the concentration available are not the same number.

What the two changes do

Substituting arginine at position 3 reduces affinity for the binding proteins. The N-terminal extension adds to the same effect. The result is an analogue that stays comparatively free in the presence of binding proteins.

That is why LR3 appears so often in cell culture work: it makes the exposure more predictable than native IGF-1 in the same dish.

The cost of the modification

It is not IGF-1. It is an analogue with deliberately altered binding behaviour, so results obtained with it describe the analogue. Where binding-protein interaction is part of the biology under study, the modification removes the thing being studied.

Reporting a result as "IGF-1 did X" when the material was LR3 is an attribution the experiment does not support.
