Three acidic residues against one lysine, and what that does to the salt form and the solution

At neutral pH BPC-157 has two aspartate and one glutamate side chains deprotonated and negative, a carboxyl terminus negative, one lysine positive and an amino terminus positive: a net charge of about minus two. The isoelectric point is around pH 4. The peptide is highly soluble in water across the useful pH range. In the acidic conditions of purification only the two basic sites carry trifluoroacetate, so counterion mass is a smaller fraction of gross mass than for a basic peptide such as MOTS-c. Dissolved in the unbuffered water of the pack, it makes the solution slightly acidic.

Charge and isoelectric point

Four acidic groups and two basic groups. Below about pH 4 the acids protonate and the peptide becomes net positive; above, it is net negative and increasingly so. Near the isoelectric point, around 4, solubility would be lowest, but even there a small polar peptide with no hydrophobic core dissolves readily. There is no practical solubility problem with this molecule.

The acidic cluster at residues 10 and 11 is a candidate site for metal binding, which the GHK-Cu and GLOW pages discuss for copper.

Counterions

Trifluoroacetate pairs with positive charges in the solid. BPC-157 has two, so at most two counterions — about 226 daltons on 1,419, or 16 per cent at the maximum. In practice the acidic groups may carry sodium or be protonated, and the certificate states the counterion content measured. Net peptide content for an acidic peptide is typically higher than for a basic one of similar size.

Acetate exchange reduces the figure further where performed.

In the blend

The two components have opposite charge character: BPC-157 net negative, the lysine-rich thymosin fragment net positive. In solution they can associate electrostatically, which is a property of the mixture that neither has alone and one more reason the blend is not the sum of its parts.

The BPC-157 + TB500 blend is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for each component of the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
