Why BPC-157 cannot be quantified by the simplest protein method, and what is used instead

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
The 280 nanometre absorbance used to quantify proteins comes from tryptophan and tyrosine side chains. BPC-157 contains neither, and no phenylalanine either, so a solution of it is transparent at 280 and a spectrophotometer reading there reports nothing. It absorbs only through its peptide bonds, below 230 nanometres, where many other substances — including the benzyl alcohol of bacteriostatic water — also absorb. Concentration is therefore established by chromatography against a standard, by amino acid analysis, or by weighing, not by direct absorbance.

The chemistry

Peptide bonds absorb strongly around 190 to 220 nanometres; aromatic rings add a weaker, distinct band near 280 that other components rarely share. A peptide with aromatics can be measured at 280 against a near-zero background; one without can be measured only at the lower wavelengths, where the background is everything. BPC-157 is in the second group, as are GHK-Cu and most short peptides.

MOTS-c, with a tryptophan and two tyrosines, is in the first group and its page sets out the calculation.

What is used instead

Reversed-phase chromatography with detection at 214 against a calibration curve of a reference of known content, which is the routine method. Amino acid analysis, which hydrolyses the peptide and quantifies its residues, for assigning the content of a reference. Gravimetric content from the certificate — net peptide content times gross mass — for a vial as supplied.

None of these is a direct reading on a solution the way 280 absorbance is.

Why it is noted

Because the blend contains one component that cannot be read at 280 and one, the thymosin fragment, that may have a single phenylalanine and still reads poorly. Direct absorbance is not a route to the concentration of either in this product. This is an analytical property, not a procedure.

The BPC-157 + TB500 blend is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for each component of the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
