A worked description of what a certificate chromatogram for the blend shows, peak by peak

On a reversed-phase gradient at 214 nanometres, the earlier of the two main peaks is BPC-157, polar and acidic; the later is the thymosin fragment, longer and lysine-rich. Small peaks just before BPC-157 are its isoaspartyl forms; small peaks around the fragment are its deletion sequences and any deamidated forms; a late peak may be an incompletely deprotected adduct of either. The composition ratio is the ratio of the two main-peak areas corrected for the response of each at 214, which differs because the two peptides have different numbers of peptide bonds. A flat baseline between the main peaks indicates that nothing co-elutes there.

Retention is by hydrophobicity. BPC-157, with four prolines, three acidic residues and no aromatics, has little to hold it and elutes early. The fragment, with hydrophobic residues around the actin-binding motif and several lysines ion-paired with trifluoroacetate, elutes later. The order is established by injecting each alone and is stated on the certificate.

The small peaks

Assigned by mass in the method development and recognised by retention time thereafter. For BPC-157, the isoaspartyl shoulder and deletion sequences. For the fragment, deletions, deamidation and oxidation if its sequence contains a susceptible residue. Each is reported under its parent. An unassigned peak above a threshold is investigated.

The sum of a component main peak and its assigned impurities is the denominator for that component purity.
