BPC-157 is a synthetic pentadecapeptide derived from a sequence found in human gastric juice, studied almost entirely by one research group in rodent injury models. TB500 corresponds to the active region of thymosin beta-4, an actin-binding peptide studied for its role in cell migration. They are read together because both literatures concern tissue repair, but they are separate bodies of work with different mechanisms and different weight of evidence.

The great majority of the published work comes from Predrag Sikiric and colleagues in Zagreb, across several decades and a wide range of rodent models — gastrointestinal lesions, tendon and ligament injury, muscle, nerve and vascular work. The mechanistic strand cited most often outside that group concerns angiogenesis and nitric-oxide pathway signalling.

The volume of papers is genuinely large. It is also concentrated in animal models and in one research group, and independent replication in humans is thin. A long citation list is not the same thing as a robustly reproduced clinical effect, and anyone evaluating this compound should hold both facts at once.

TB500: a fragment of a well-characterised protein

Thymosin beta-4 is a 43-amino-acid peptide present in most mammalian cells and among the most abundant actin-binding proteins described. TB500 is a synthetic fragment corresponding to its active region.

The protein was characterised largely through the work of Allan Goldstein and colleagues. Research since has examined it in corneal and dermal repair models and in cardiac tissue, with mechanistic interest centring on G-actin sequestration and the regulation of cell motility.

Unlike BPC-157, thymosin beta-4 has been studied by multiple independent groups and has reached clinical trials in some indications. The evidence base has a different shape, even though the preclinical framing is similar.

Why one carton
