Myostatin, follistatin, and what the genetics actually showed

Myostatin, also called GDF-8, was described by Alexandra McPherron and Se-Jin Lee in 1997 as a negative regulator of skeletal muscle mass; mice lacking it grew substantially more muscle. Follistatin binds myostatin, which is why the two are discussed together.

What the 1997 work reported

McPherron and Lee identified GDF-8 and showed that mice in which the gene was disrupted developed markedly greater skeletal muscle mass. The phenotype was striking enough that the same loss-of-function was later recognised in cattle breeds and in other species.

That is a genetics result: remove the gene from the start of development, and the animal grows differently. It is strong evidence that myostatin restrains muscle growth.

Where follistatin enters

Follistatin binds myostatin among its other targets, so it can reduce the amount available to signal. That connects a binding protein to a well-characterised growth restraint, and it is the reason the two names appear together in almost every summary.

The connection is real and it is mechanistic. It is also where the honest account stops.

The step the marketing takes and the literature does not

A lifelong genetic knockout in a mouse is not the same claim as an exogenous protein added to an adult animal, and neither is a claim about people. The distance between those three statements is where most follistatin marketing lives.

What a laboratory can test is a specific, bounded question in a defined model. That is worth doing, and it is not the same as the claim printed on a supplement page.
