The copper enzyme that cross-links collagen and elastin, and the route by which a copper peptide could reach it

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Lysyl oxidase is a secreted enzyme that oxidises specific lysine residues on collagen and elastin to aldehydes, which then condense spontaneously into the covalent cross-links that give the fibres their tensile strength and resistance to degradation. It requires a copper ion and a derived quinone cofactor, and without copper the enzyme is made but inactive. Copper deficiency in animals and in Menkes disease in humans produces fragile vessels, weak bone and loose skin through this enzyme. The connection to GHK-Cu is a hypothesis: a peptide that delivers copper to fibroblasts could support lysyl oxidase activity and so the quality of newly made matrix, not only its quantity.

The enzyme

Lysyl oxidase and its four relatives act outside the cell on assembled collagen fibrils and elastin. The aldehyde products react with neighbouring lysines and with each other to form the pyridinoline and desmosine cross-links that mature matrix contains. Cross-linking increases with age and is part of why old collagen is stiff and resistant to turnover. The enzyme is also implicated in fibrosis and in tumour matrix stiffening.

It is one of about a dozen copper enzymes in humans, alongside ceruloplasmin, cytochrome c oxidase and superoxide dismutase.

The copper connection

The enzyme acquires copper inside the cell before secretion, through the transporter ATP7A in the secretory pathway. Cells therefore need an adequate copper supply to make active enzyme, and copper-deficient fibroblasts secrete inactive protein. A copper complex that cells take up would feed this pathway. Whether GHK-Cu does so in any tissue, and whether that matters beside dietary copper, is not established; the pathway is.

This is one of the routes the copper-delivery hypothesis proposes.

Why it is here

Because the biology of copper in connective tissue is the reason a copper peptide was thought worth studying in that tissue. It describes biochemistry and no use of the pack.

GHK-Cu is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
