A classical route to peptide content, and its limits for GHK-Cu

Published 24 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Total nitrogen in a sample, measured by combustion or digestion methods, can be converted to peptide content using the nitrogen fraction of the peptide. GHK is nitrogen-rich — histidine contributes three nitrogens and lysine two beyond the backbone — so its conversion factor is specific to it and cannot be borrowed from a generic protein value. Trifluoroacetate counterion contributes no nitrogen; acetate contributes none; so the method is unaffected by salt form.

The methods

Combustion analysis burns the sample and measures nitrogen gas; digestion methods convert nitrogen to ammonia and titrate it. Both give total nitrogen by mass. Neither distinguishes peptide nitrogen from any other nitrogen-containing impurity.

They are rapid and well established.

The conversion factor

Peptide content equals measured nitrogen divided by the nitrogen fraction of the pure peptide. For GHK, with its imidazole and amine nitrogens, that fraction is higher than for an average protein, and using a generic factor would understate content. The factor is calculated from the sequence.

Copper does not contribute nitrogen and does not interfere.

Where it sits among the methods

A useful cross-check against amino acid analysis and the difference method. Agreement among all three is strong evidence that a stated peptide content is right.
