Human growth hormone binds a second receptor that most growth hormones ignore, and zinc is the reason

Human growth hormone activates not only its own receptor but also the prolactin receptor, giving it lactogenic as well as somatogenic activity; non-primate growth hormones lack this. Cunningham, Bass, Fuh and Wells reported in Science in 1990 that binding to the prolactin receptor requires a zinc ion coordinated between histidine and glutamate residues on the hormone and a histidine on the receptor. The dual activity matters for interpreting bioassays that use prolactin-responsive cells and for the physiology of the hormone in pregnancy and lactation.

The 1990 finding

Binding of human growth hormone to the extracellular domain of the prolactin receptor was found to depend on zinc at physiological concentration and to be abolished by chelation. Mutagenesis located the zinc ligands: histidine 18, histidine 21 and glutamate 174 on the hormone, and a histidine on the receptor. Binding to the growth hormone receptor needed no zinc. A single metal ion therefore switches one hormone between two receptors.

It was an early example of metal-dependent protein-protein recognition in a hormone system.

Consequences

In vivo, human growth hormone has prolactin-like effects on the breast and reproductive tissues that animal hormones lack, and these appear among its known effects at high levels. In vitro, the widely used Nb2 cell assay responds through the prolactin receptor, so it measures lactogenic potency; assays using cells engineered with the growth hormone receptor measure somatogenic potency. The two usually agree for intact hormone and can diverge for variants.

Placental lactogen and prolactin, the other members of the family, bind the prolactin receptor without zinc.

Why it is here

Because it is part of what is established about the receptor pharmacology of the molecule in the pack, and because a certificate potency figure depends on which receptor the assay used.
