Does growth hormone act directly or through IGF-1? What the experiments said

In 1957 Salmon and Daughaday showed that growth hormone added to cartilage in vitro did nothing, while serum from hormone-treated animals stimulated it, and proposed an intermediary they called sulfation factor, later somatomedin and then IGF-1. In 1999 two groups, Yakar and colleagues and Sjögren and colleagues, deleted the IGF-1 gene in mouse liver only; circulating IGF-1 fell by three quarters and the mice grew normally. The revised view is that locally produced IGF-1 and direct growth hormone actions both contribute, with liver-derived IGF-1 being less necessary for growth than the original hypothesis held.

The original experiment

Rat cartilage in culture takes up sulfate as it makes matrix. Growth hormone added directly did not increase uptake; serum from animals given growth hormone did. The simplest explanation was a circulating mediator made in response to the hormone. Published in the Journal of Laboratory and Clinical Medicine in 1957.

The factor was purified over the following two decades and shown to be identical to IGF-1.

The 1999 test

Liver makes most of the IGF-1 in blood. Deleting the gene there, using a liver-specific recombinase, removed the endocrine supply while leaving local production elsewhere intact. The mice reached normal size, which was not what a strictly endocrine hypothesis predicted. Both papers appeared in the Proceedings of the National Academy of Sciences.

Later work showed effects on other systems in those mice, so circulating IGF-1 is not without function; it is not required for linear growth.

The current framing

Growth hormone acts directly on some tissues, induces IGF-1 locally in many, and induces liver IGF-1 that acts as a hormone. The three are not alternatives. The history is a clear example of a correct observation generating a hypothesis that was then narrowed by a sharper experiment.
