A growth factor that crosses into the brain, and the studies linking it to exercise and new neurons

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
IGF-1 is transported across the blood-brain barrier by a receptor-mediated process and is also synthesised within the brain, with receptors on neurons and glia throughout. Carro, Torres-Alemán and colleagues reported in 2000 that exercise increased the uptake of circulating IGF-1 into the rodent brain, and Trejo and colleagues reported in 2001 that blocking circulating IGF-1 abolished the increase in hippocampal neurogenesis that exercise produces. Human data are observational: lower IGF-1 associates with poorer cognition in some ageing cohorts and not others, and no trial has shown cognitive benefit from raising it.

Entry and production

The barrier endothelium expresses IGF-1 receptors and transports the hormone by transcytosis, with the choroid plexus a second route. Neurons and astrocytes make IGF-1 locally, especially during development, when the IGF1 gene deletion case showed its necessity for brain growth and hearing. In the adult brain it is studied for neuroprotection, synaptic plasticity and the regulation of amyloid clearance.

The brain is one of the tissues where local and circulating IGF-1 both contribute.

The exercise studies

Running rodents took up more circulating IGF-1 into the brain, and the hormone accumulated in neurons activated by the exercise. Antibodies that neutralised circulating IGF-1 prevented the exercise-induced birth of new neurons in the hippocampus and the protective effect of exercise against brain injury. These studies made circulating IGF-1 a candidate mediator of the cognitive effects of exercise.

Later work added other mediators, so IGF-1 is one of several.

The human picture

Cohort studies relating IGF-1 to cognition or dementia risk are inconsistent in direction. The Laron cohort, with lifelong low IGF-1, has not shown cognitive deficit. No interventional data exist. This page describes rodent mechanism and human association, and describes no use of the pack.
