Where a growth factor goes when there is very little of it in a lot of liquid

Proteins adsorb to surfaces through hydrophobic and electrostatic contact, and the amount adsorbed is roughly fixed by the surface area rather than by the concentration. At milligram-per-millilitre concentrations the loss is negligible; at nanograms per millilitre it can be most of the protein. IGF-1 LR3, with a hydrophobic core and a basic isoelectric point, adsorbs readily to polystyrene and glass. The cell culture literature addresses this by including a carrier protein such as albumin in dilute solutions, which saturates the surfaces. An experiment that omits it may be measuring a lower concentration than it believes.

The physics

A surface has a finite number of binding sites. A dilute solution gives them up a fixed amount of protein; what remains in solution is the difference. The fraction lost therefore rises as concentration falls. Polystyrene is strongly adsorptive; polypropylene less so; glass binds basic proteins through its negative surface. Low-binding plasticware reduces but does not eliminate the effect.

This is standard surface chemistry and applies to every protein, more or less.

What the literature does

Published methods for growth factors in culture typically specify a carrier protein in the diluent — bovine or human serum albumin at a fraction of a per cent — so that the surfaces are coated with the carrier and the growth factor stays in solution. Media manufacturers formulate accordingly. Where a defined, protein-free medium is required, recombinant albumin or other blocking agents are used.

These are descriptions of published practice, not instructions.

Why it is on this page

Because a 1 mg vial becomes very dilute solutions in use, and adsorption is one of the reasons two laboratories can report different results from the same lot. It is a property of the molecule and of surfaces, stated as such.

IGF-1 LR3 is supplied by REVIVE LAB UAE for laboratory research use only, with identity and purity documented for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
