The direct content method applied to a protein, and what the ratios confirm

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Amino acid analysis hydrolyses the protein in strong acid at elevated temperature, breaking all peptide bonds, then separates and quantifies the free amino acids against standards. For IGF-1 LR3 the expected composition follows from the 83-residue sequence, and the measured ratios confirm it; the total gives protein content independent of water and counterion. Tryptophan is destroyed by acid hydrolysis and cysteine must be derivatised first, so those are measured separately or omitted, and asparagine and glutamine appear as their acids.

The method

A weighed sample is hydrolysed, typically in hydrochloric acid at around 110 degrees for a day, under vacuum or inert gas. The hydrolysate is analysed by ion-exchange chromatography with post-column derivatisation or by pre-column derivatisation and reversed phase. Each amino acid is quantified, and the sum, corrected for the residues not recovered, is the protein content.

The method is slower than spectrophotometry and more accurate.

What the ratios confirm

The sequence predicts, for example, six cysteines, one methionine and defined counts of each other residue. Measured ratios close to prediction confirm the composition; a deviation points to a truncation, a wrong sequence or a contaminant. For a recombinant protein this is a second identity check independent of mass.

The extension adds its own residues to the expected counts, so the analogue and native IGF-1 give different compositions.

On the certificate

Protein content by amino acid analysis, often used to assign the content of a reference lot against which routine lots are measured by absorbance or chromatography. A 1 mg vial contains the stated mass of lyophilised material; this method says how much of it is protein.
