How a disulfide-rich protein is recovered from bacteria that cannot fold it

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Expressed at high level in E. coli, IGF-1 LR3 misfolds and aggregates into dense inclusion bodies, because the bacterial cytoplasm is reducing and cannot form disulfides. The bodies are isolated by cell lysis and centrifugation, dissolved in a strong denaturant such as guanidine hydrochloride with a reducing agent, and the unfolded protein is then diluted into a buffer containing a redox pair, typically oxidised and reduced glutathione, that lets disulfides form and reshuffle until the most stable arrangements are reached. The native form and the swap isomer both emerge, and chromatography separates them.

Why inclusion bodies

Rapid synthesis of a protein that cannot reach its native state leaves partly folded chains with exposed hydrophobic surfaces, which associate and precipitate. For disulfide-containing proteins this is the rule rather than the exception in E. coli. Inclusion bodies are in one sense convenient: they are dense, relatively pure and protect the protein from proteases.

The 13-residue extension of LR3 was originally part of making this expression workable.

The refolding step

Dilution lowers protein concentration so that intramolecular folding outpaces intermolecular aggregation. The redox buffer sets a mild oxidising potential under which thiols pair, mispair and exchange until stable disulfides form. Temperature, pH, time and additives are optimised for each protein. Yield of native protein is typically a fraction of the input, and the balance is isomers and aggregates.

This step is where the swap isomer arises and is why it must be controlled downstream.

What follows

Purification by ion-exchange and reversed-phase chromatography, with the latter resolving the disulfide isomers; buffer exchange; and lyophilisation. The certificate reports the outcome. This page describes a manufacturing process in general terms.
