The receptor arithmetic that explains why one replaced the other

Insulin binds the IGF-1 receptor with roughly a hundredfold lower affinity than IGF-1 does. In serum-free media it was therefore used at microgram-per-millilitre concentrations, hundreds of times physiological, so that enough reached the IGF-1 receptor to drive proliferation. IGF-1 LR3 binds that receptor at near-native affinity and escapes the binding proteins the cells secrete, so it achieves the same effect at nanogram-per-millilitre concentrations. Media developers adopted it for that reason, and it also removed insulin, an animal-derived or recombinant pharmaceutical, from the formulation.

Why insulin worked at all

The insulin and IGF-1 receptors are close relatives and each binds the other ligand weakly. At high enough concentration insulin occupies IGF-1 receptors and signals through them, and also signals through its own receptor, which many cell lines express. The effect was real but inefficient, and the concentrations needed were far from anything physiological.

Insulin also degrades in culture medium over days, requiring replenishment.

Why the analogue displaced it

Direct, high-affinity agonism of the intended receptor; no loss to binding proteins; stability in medium; and a defined, recombinant, animal-origin-free source. The amount needed fell by two to three orders of magnitude. For large-scale production of therapeutic proteins, where media cost and definition matter, that was decisive.

Some media retain insulin for its own receptor effects and add the analogue for the IGF-1 receptor.

The boundary

This page describes a substitution made in the formulation of commercial culture media and the receptor pharmacology behind it. It describes no use of the pack.

IGF-1 LR3 is supplied by REVIVE LAB UAE for laboratory research use only, with identity and purity documented for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
