A receptor tyrosine kinase built like the insulin receptor

The IGF-1 receptor complementary DNA was reported by Ullrich and colleagues in the EMBO Journal in 1986, a year after the same group cloned the insulin receptor. It is a disulfide-linked tetramer of two extracellular alpha subunits that bind ligand and two membrane-spanning beta subunits with intracellular tyrosine kinase domains. The two receptors share about seventy per cent identity in the kinase domain and bind each other ligands weakly; the hybrid receptors formed from one half of each are a consequence of the similarity.

The architecture

A single precursor is cleaved into alpha and beta; two alpha-beta halves are joined by disulfides into the mature receptor. Ligand binds across the two alpha subunits, and the conformational change activates the two kinase domains, which phosphorylate each other and then docking proteins, chiefly the insulin receptor substrates.

Unlike most receptor tyrosine kinases, it is a covalent dimer before ligand arrives.

Same family, different ligand preference

The IGF-1 receptor binds IGF-1 with about a hundredfold higher affinity than insulin, and the insulin receptor the reverse. IGF-2 binds both. The structural basis lies in the ligand-binding alpha subunits, which differ more than the kinase domains do. The downstream machinery is largely shared, which is the basis of the crosstalk discussed elsewhere on this site.

The receptor is expressed on nearly every cell type, which is why IGF-1 acts so broadly.

Why it matters for LR3

IGF-1 LR3 binds this receptor with affinity close to that of native IGF-1 — the modifications were selected to spare receptor binding while reducing binding-protein affinity. The receptor is therefore the same target; what differs is how much free ligand reaches it.

IGF-1 LR3 is supplied by REVIVE LAB UAE for laboratory research use only, with identity and purity documented for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
