One tryptophan and two tyrosines make MOTS-c measurable by absorbance, which most short peptides are not

Absorbance at 280 nanometres comes almost entirely from tryptophan and tyrosine. Using the residue values of Pace and colleagues from 1995 — about 5,500 per molar per centimetre for tryptophan and 1,490 for tyrosine — MOTS-c has a calculated molar absorptivity near 8,480. A solution of known path length therefore gives concentration directly from absorbance. The method is simple and non-destructive, measures total peptide including oxidised forms, and is unavailable for GHK-Cu, BPC-157 and other catalogue peptides that lack aromatic residues.

The calculation

Molar absorptivity of a peptide at 280 is the sum over its tryptophans, tyrosines and disulfide bonds. MOTS-c has one, two and none, giving about 5,500 plus 2,980, or 8,480. Concentration in moles per litre is absorbance divided by that figure and by the path length in centimetres. At 2,174 grams per mole this converts to mass concentration.

The values are for the residues in a folded protein; in a small unstructured peptide they are close but not exact, and a few per cent error is normal.

What the method sees

Every molecule carrying the aromatic residues, including oxidised peptide, truncated peptide that retains them, and any aromatic impurity — benzyl alcohol from bacteriostatic water absorbs in this region and must be blanked. It reports total aromatic content, not purity. Chromatography reports purity.

Tryptophan oxidation reduces absorbance at 280 and shifts it, so a heavily oxidised sample reads low.

Why it is noted

Because it is an analytical property that distinguishes this peptide from most of the catalogue and because the blank requirement with the supplied diluent is a practical fact worth stating. It is not a procedure for any purpose.

MOTS-c is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
