What the 2015 paper measured in ageing mice, and what later cohorts reported in people

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Lee and colleagues reported in 2015 that MOTS-c in skeletal muscle and in plasma declined with age in mice, measured by immunoassay. Human studies since have described lower circulating levels in older than younger adults, and the 2021 exercise study measured it in muscle and plasma of young men. The pattern is consistent across the reports that exist, with the caveat that immunoassays for short peptides vary between laboratories and that absolute values are not comparable across studies.

The mouse data

Young and old mice were compared for MOTS-c content in muscle by western blot and in plasma by enzyme-linked immunoassay; both were lower in old animals. The finding was part of the case for the peptide as a metabolic regulator whose loss accompanies ageing, parallel to earlier reports of humanin declining with age.

It is an association in a cross-sectional comparison, not a demonstration of cause.

The human data

Several groups have measured plasma MOTS-c in cohorts of different ages and in disease states, generally reporting lower levels with age and in some metabolic conditions. The exercise study of 2021 found levels rising in muscle and plasma after a bout of exercise in young men. Sample sizes are small and methods differ.

No study has measured levels across the lifespan in one population with one method.

The measurement caveat

A 16-residue peptide at low concentration in plasma is hard to measure specifically; antibodies can cross-react with fragments or precursors, and oxidised forms may or may not be detected. Mass spectrometric methods are emerging. The direction of the age effect is reasonably consistent; the magnitude is not established.

MOTS-c is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
