What the energy sensor is made of, how it is switched on, and what it does once it is

AMP-activated protein kinase is a heterotrimer: a catalytic alpha subunit, a scaffolding beta subunit, and a regulatory gamma subunit that binds AMP, ADP and ATP at adenine-nucleotide sites. When AMP rises relative to ATP, binding to gamma promotes phosphorylation of the alpha subunit by the upstream kinase LKB1 — identified in 2003 by the Hardie and Carling groups among others — and protects that phosphorylation from removal. Active AMPK phosphorylates acetyl-CoA carboxylase to increase fat oxidation, inhibits mTOR to reduce protein synthesis, promotes GLUT4 translocation, and activates PGC-1 alpha to increase mitochondrial biogenesis. It is the common node that AICAR, metformin, exercise and, by the 2015 proposal, MOTS-c all reach.

Structure and activation

The gamma subunit has four nucleotide-binding sites of which three are used; AMP and ADP binding changes the conformation so that threonine 172 on alpha is phosphorylated and stays phosphorylated. Calcium, through a second upstream kinase, activates it independently of energy state. Drug-binding sites on the alpha-beta interface are the targets of newer direct activators. There are twelve possible subunit combinations with tissue-specific distribution.

The Hardie laboratory in Dundee has been the main source of this biochemistry since the 1980s.

What it does

In one sentence: it turns off ATP-consuming anabolic processes and turns on ATP-generating catabolic ones. Fatty acid synthesis down, fatty acid oxidation up; protein synthesis down; glucose uptake up; mitochondrial biogenesis up over longer periods; autophagy up. In liver it reduces gluconeogenesis and lipogenesis. The phenotype of sustained activation resembles that of exercise training, which is the origin of the exercise-mimetic idea.

Chronic activation is not uniformly beneficial; it also suppresses growth and some repair processes.

Why it is here

Because the MOTS-c mechanism, as proposed, ends at this enzyme, and a reader should know what the enzyme is. This is biochemistry, stated as such.

MOTS-c is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
