The specific metabolic route the 2015 paper proposed, step by step

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
In the 2015 Cell Metabolism paper, metabolomic profiling of cells exposed to MOTS-c showed depletion of folate-cycle intermediates and of methionine-cycle products, together with accumulation of 5-aminoimidazole-4-carboxamide ribonucleotide, AICAR, a purine-synthesis intermediate that is also a known activator of AMP-activated protein kinase. The authors proposed that the peptide inhibits the folate cycle, that purine synthesis stalls at the step needing a folate cofactor, that AICAR builds up, and that AICAR activates AMPK, which then increases glucose uptake and fatty acid oxidation. The metabolite changes and AMPK activation were measured; the direct molecular target was not identified.

The folate and methionine cycles

One-carbon metabolism moves methyl groups through folate derivatives and uses them to regenerate methionine from homocysteine and to build purines. Blocking the folate side depletes methionine-cycle output and starves purine synthesis of the one-carbon unit it needs at the AICAR transformylase step, so AICAR accumulates. These are textbook pathways, and the profile reported was consistent with a block at that point.

The same AICAR accumulation is why the folate antagonist methotrexate activates AMPK.

AICAR and AMPK

AICAR is taken up by cells and phosphorylated to ZMP, an AMP mimic that binds the regulatory subunit of AMPK and activates it. AICAR has been used as a pharmacological AMPK activator for decades, which is why its accumulation offered a ready explanation for the AMPK activation observed. AMPK in turn promotes glucose uptake, fatty acid oxidation and mitochondrial biogenesis.

AMPK activation by MOTS-c was shown by phosphorylation in cells and in mouse muscle.

What remains inferred

The enzyme or process MOTS-c acts on to block the folate cycle has not been identified. The chain from peptide to AMPK is a proposed mechanism supported by metabolomics and consistent with known biochemistry, not a demonstrated binding event. The 2018 nuclear-translocation work added a second, independent mode of action.
