The technique that generated the MOTS-c mechanism, what it measures, and how a hypothesis comes out of it

Untargeted metabolomics extracts small molecules from cells or tissue and separates and detects them by liquid chromatography coupled to high-resolution mass spectrometry, identifying hundreds to thousands of features and comparing their abundance between conditions. In the 2015 MOTS-c work it showed depletion of folate-cycle and methionine-cycle intermediates and accumulation of AICAR in treated cells, which pointed to a block in one-carbon metabolism. The method is powerful for finding what changed and silent on why: it produces a map of altered metabolites from which a hypothesis is drawn, and the hypothesis then needs targeted experiments. The MOTS-c folate hypothesis has had some such follow-up and still lacks an identified target.

How it works

Cells are quenched rapidly to freeze metabolism, extracted with cold solvent, and the extract run through chromatography into a mass spectrometer that records the exact mass of everything that elutes. Features are matched to databases by mass and fragmentation and compared statistically between groups. Pathway analysis then asks which pathways are over-represented among the changed metabolites.

Identification is confident for well-known metabolites and tentative for many others.

What it can and cannot show

It can show that a pathway is perturbed and in which direction. It cannot show which enzyme was affected, whether the change is cause or consequence, or whether the peptide acted on the pathway directly. For MOTS-c the AICAR accumulation was a strong lead because AICAR is a known AMPK activator; the lead has not yet been traced to a specific enzyme inhibition.

Many metabolomics-derived hypotheses in the literature remain at this stage for years.

Why it is here

Because the status of the MOTS-c mechanism — a well-supported hypothesis without an identified target — follows from the method that produced it, and a reader should understand both. It describes no use of the pack.

MOTS-c is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
