An HPLC chromatogram plots detector response against retention time. Purity is reported as the main peak area as a percentage of total integrated peak area. Reading the trace rather than only the number shows where the remaining percentage sits and whether the integration was reasonable.

What the axes mean

Time runs along the bottom: compounds leave the column in order of how strongly they interact with it, so retention time is an identity clue under fixed conditions. Detector response runs up the side, usually UV absorbance at a wavelength where peptide bonds or aromatic residues absorb.

A peak is a compound leaving the column. Its area is proportional to how much arrived at the detector, which is why purity is quoted as an area percentage rather than a height one.

The number and what it leaves out

Ninety-eight per cent purity means the main peak carries 98 per cent of the integrated area. It does not say what the other two per cent is, and two per cent of one impurity is a different situation from two per cent spread across six.

Nor does an area percentage account for anything the detector cannot see. A species that does not absorb at the chosen wavelength contributes nothing to the total and is therefore invisible to the calculation — one of the reasons mass spectrometry is run alongside rather than instead.

What to look at on the trace itself

A flat, quiet baseline, a symmetrical main peak rather than one with a shoulder or a tail, small peaks that are resolved from the main one rather than riding on it, and integration marks placed where the peak actually starts and ends. A peak integrated generously across a shoulder inflates the number without any chemistry changing.

Also check that the method conditions are stated: column, mobile phase and gradient, detection wavelength. A purity figure without its method is not reproducible and not comparable with anyone else's figure.

The complex adds one consideration

GHK-Cu is a metal complex, and chromatography of metal complexes can be affected by the mobile phase and by any chelating component in it. This is why identity is confirmed by mass spectrometry as well, and why the analytical questions for a copper peptide are not identical to those for a plain peptide.
