Serum albumin binds copper(II) with high affinity at its N-terminus and is a principal carrier of exchangeable copper in plasma. Free histidine binds it too, and chelators such as EDTA are present in many buffers. Any copper complex added to such a system is competing for its own metal.

Albumin is the obvious competitor

Albumin is abundant and binds copper(II) tightly at a specific N-terminal site. In serum-containing culture medium it is present in quantity, which means an added copper complex enters a solution that already has a strong copper acceptor in it.

Whether the copper stays on the peptide, transfers to albumin, or reaches some equilibrium between them is a question about relative affinities and concentrations — not something that can be assumed from what was added to the tube.

Buffers are not neutral either

EDTA appears in many buffers and media formulations and is a strong chelator. Free histidine binds copper. Even the buffer species itself can coordinate a metal in some systems.

A result obtained in one buffer is therefore not automatically reproducible in another, and a methods section that does not state the full composition has left out something that matters for a metal complex in a way it would not for a plain peptide.

What this means for reading a paper

Ask what was in the medium. Serum or serum-free, which buffer, whether any chelator was present. For copper work these are not housekeeping details; they are part of the experiment.

It is also why in vitro copper results can be harder to reproduce between laboratories than the apparent simplicity of the system suggests.
