The slow degradation that changes charge but barely changes mass

Published 24 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Deamidation converts an asparagine or glutamine side-chain amide to a carboxylic acid, adding a negative charge and shifting mass by roughly one dalton. It proceeds slowly in solution, faster at elevated pH and temperature, and is harder to detect than oxidation because the mass change is small. It is a standard degradation pathway considered in peptide stability work.

The reaction

Asparagine deamidates through a cyclic succinimide intermediate that opens to give either aspartate or isoaspartate — the latter a rearranged form that can alter the peptide's structure and activity. Glutamine deamidates more slowly by a related route.

Rate depends strongly on the neighbouring residue, on pH and on temperature, which is why sequence context and storage conditions both matter.

Why it is hard to see

A one-dalton mass shift is within the isotope envelope of a peptide of this size, so it does not stand out in a routine mass spectrum the way a 16-dalton oxidation does. The charge change does shift retention on some chromatographic methods, and dedicated methods exist to resolve deamidated forms.

A certificate that reports purity by a method insensitive to deamidation has not excluded it, and the better certificates say what their method can and cannot resolve.

What storage does

Deamidation needs water. Lyophilized material is far less susceptible than solution, and solution at low temperature far less than at room temperature. This is among the reasons a reconstituted peptide has a limited life even when nothing visible has changed.

How long a given solution remains within specification is a stability question answered by measurement in that system, not by a general rule.
